Abstract: This paper describes an easy and simple method to perform yeast cell experiment on a flow cytometry chip. We are able to observe the influx-hydrolysis-efflux phenomenon of a fluorogenic substrate on yeast cells. As a study model, the fluorogenic substrate fluorescein diacetate (FDA) diffused into the yeast cell, and FDA was hydrolyzed in the cytosol to produce fluorescein. Then, fluorescein was effluxed from the yeast cell.
